چکیده

Using 3-dimensional cultured human skin and epidermal models, an alternative to skin irritation testing have been developed. In the ECVAM (European Centre for the Validation of Alternative Methods) validation protocol, hazardous chemicals have been identified using these models. We considered it likely that these models would also be useful for examining the dose-response of the toxicity of a chemical. Therefore, we used the epidermal model (Labcyte, Japan Tissue Engineering Co. Ltd.) to evaluate the dose-response of cytotoxicity and compared the findings with those for irritancy by human patch testing of 4 chemicals: sodium lauryl sulfate, benzethonium chloride, nonanoic acid and propylene glycol. The cytotoxicity of the chemicals was found to be stronger than the irritancy shown by human patch data. Difference ratios ranging from 1.4 to 52.7 were found between irritancy and EC50(concentration causing a 50% reduction in the MTT assay compared to the untreated control value) in LabCyte for 4 chemicals and solvents. We consider that these models are useful for evaluating the dose-response of skin irritancy, and could be used to establish a database for risk assessment of chemicals.

Assessment of the Dermal and Ocular Irritation Potential of Lomefloxacin by Using In Vitro Methods

The evaluation of eye and skin irritation potential is essential to ensuring the safety of human in contact with a wide variety of substances. Despite this importance of irritation test, little is known with respect to the irritation potency of lomefloxacin, a fluoroquinolone antibiotic, which has been known to cause phototoxicity with an abnormal reaction of the skin. Thus, to investigate the ...

Hallmarks of atopic skin mimicked in vitro by means of a skin disease model based on FLG knock-down.

Loss-of-function mutations in the filaggrin gene (FLG) are a strong predisposing factor for atopic dermatitis, although their relevance to the disease pathomechanism needs further elucidation. The generation of an in vitro model of atopic skin would not only permit further evaluation of the underlying pathogenetic mechanisms and the testing of new treatment options, but would also allow toxicol...

[Development of alternative to animal experiment in evaluation of skin irritation caused by alcohol-based hand rubs].

Alcohol-based hand rubs are widely used for infection control in clinical practice. However, it is known that frequent use of the alcohol-based hand rubs may cause skin irritation. To predict the skin irritation in human, animal experiments are quite useful. Especially, the Draize Test using rabbits is suitable for this purpose because their skin is highly sensitive. On the other hand, the deve...

Assessment of phototoxicity, skin irritation, and sensitization potential of polystyrene and TiO2 nanoparticles

The human skin equivalent model (HSEM) is well known as an attractive alternative model for evaluation of dermal toxicity. However, only limited data are available on the usefulness of an HSEM for nanotoxicity testing. This study was designed to investigate cutaneous toxicity of polystyrene and TiO2 nanoparticles using cultured keratinocytes, an HSEM, and an animal model. In addition, we also e...

[Construction of three-dimensional human skin model involving dendritic cells and its application to skin sensitization test].

Recently, to study an in vitro evaluation method of skin irritation and acute toxicity, many three-dimensional human skin models consisting of normal human keratinocytes and fibroblasts have been used. However, these skin models did not have any dendritic cells so were difficult to apply to an in vitro skin sensitization test. On the other hand, a single cell-culture model using normal human de...
